It is a good idea to use some sort of softening agent or spray to try and loosen the dirt beforehand and perhaps eliminate the need to use a comb altogether. If there is a disconnected duct or breach in the airway, the conditioned air will be contaminated and reduce the effectiveness of the AC production. Some rooms may be cooler than others and there may be smells or dirt inside of those rooms coming through the vents.
